The ribonuclease L (RNase L; RNASEL) gene encodes a key enzyme in the interferon induced antiviral and anti-proliferate pathway, in particular through the mobilization of type I interferons, and is thereby involved in cellular viral defense, single-stranded RNA cleavage and tumor suppressor activities such as stress mediated apoptosis, cell proliferation and regulation of protein synthesis.
RNASEL SNPs associated with cancer risk include:
- rs486907, known as R462Q
- rs3738579, a SNP in the 5' UTR potentially associated with multiple cancers
- A SNP lacking an rs# in dbSNP, known as E265X, associated with an 11 year earlier onset of prostate cancer in predisposed families
