By itself this SNP is associated with a 2x-3x increased risk of developing late-onset Alzheimer's disease. However [PMID 17192785] suggests that this SNP really only serves as a proxy for the ApoE4 allele which has a larger risk increase. Since feedback from Promethease users suggests that this SNP may not be such an accurate proxy, you may wish to specifically verify your APOE allele status by checking genoset gs216, which checks both rs429358 and rs7412, as well as any other APOE alleles described on the APOE page.
